以当年产当归种子为材料,分别对种子进行去翅和不去翅处理,观察其无菌苗培养条件及愈伤组织诱导、继代培养情况。结果表明,当归种子去翅后,在1/2 MS培养基上有利于获得无菌苗,发芽率达55.4%,污染率仅为6.5%。叶柄为当归愈伤组织诱导的最佳外植体,H培养基为当归诱导愈伤组织的适宜基本培养基。诱导愈伤组织培养基为H+0.5 mg/L 2,4-D时,出愈率达90%以上;继代增殖培养基为H+0.5 mg/L 2,4-D+1.0 mg/L IAA时,增殖效果较好。

The seeds peeled and non-peeled of Angelica sinensis are used to study the influence of different hormone combinations and media on callus induction and plant regeneration. The result shows that seeds peeled on 1/2 MS culture medium to cultivate, germination rate is 55.4%, and the pollution rate is only 6.5%; Petiole are the optimal explant, and H medium are the optimal medium; the optimal medium for callus induction is H+0.5mg/L 2,4-D, and the callus forming rate could reach to 90%; the optimal medium for induction of subculture is H+0.5 mg/L 2,4-D+1.0 mg/L IAA, and the effect of proliferation is better .
