采用四唑染色法研究了染色温度、染色时间、TTC浓度对小叶黑柴胡种子生活力(染色效果)的影响,探讨测定小叶黑柴胡种子生活力的最佳方法,明确其生活力与发芽率之间的相关性。结果表明:影响小叶黑柴胡种子生活力测定值大小的因素依次为染色温度、染色时间、TTC浓度,得到生活力(X) 与发芽率(Y)线性关系为Y=2.603X-1.617(r=0.985)。小叶黑柴胡种子生活力测定的最佳条件为温度35 ℃、染色时间3 h、TTC浓度0.2%,测得的生活力与发芽率呈显著正相关。

The optimal conditions of TTC method for determinating seed viability of Bupleurum smithii Wolff var. parvifolium Shan et Y.Li is determined by means of analyzing the effects of dyeing temperature, dyeing time and TTC concentration on dyeing effects, the correlation between seed viability and germination rate for Bupleurum smithii Wolff var. parvifol ium Shan et Y. Li is determined. The result shows that the order of the factors affecting the estimated value of seed viability is dyeing temperature, dyeing time and TTC concentration, the linear relation between viability(X) and germination rate(Y) is Y=2.603X-1.617(R=0.985). The optimal conditions of TTC Method for determinating seed viability of Bupleurum smithii Wolff var. parvifolium Shan et Y. Li is dyeing temperature 35 ℃, dyeing time 3 hours and TTC concentration 0.2%, the seed viability and germination rate presented a significant positive correlation.
